Quality and Quality Control 

We guarantee the highest quality in our synthesized DNA and RNA oligos through rigorous QC testing,
including RP-HPLC, IEX-HPLC, MALDI-MS, ESI-MS, and PAGE.

All oligos are produced under ISO 13485 standards to ensure exceptional purity and reliability.

Quality Control Method
Description

ESI-MS

The preferred method for characterizing oligonucleotides over 50 bases is ESI-MS. At Ella Biotech, we use ESI-MS for all oligonucleotides over 30mers and for all those produced according to ISO 13485.

MALDI-MS

MALDI-TOF MS is the optimal choice for analyzing oligonucleotides under 50 bases, such as PCR primers.
At Ella Biotech, we use MALDI-MS as our preferred method for internal high-throughput quality control. We perform quality control checks on oligonucleotides from each production run, as well as all modified oligonucleotides and dual-labeled probes produced.

IEX-HPLC

Ion-exchange HPLC is the ideal method for separating oligonucleotides up to 40 bases by charge, effectively isolating full-length oligos from impurities, even in high-GC content sequences. At Ella Biotech, we use IEX-HPLC for both purification and quality control.

RP-HPLC

In our production, we use RP-HPLC for efficient oligonucleotide purification and quality control. This method separates molecules based on hydrophobicity, retaining full-length sequences on the column while shorter ones are washed off. With our experience, RP-HPLC achieves around 80% purity and is effective for oligonucleotides up to 50 bases.

UV Spectrophotometry

Ultraviolet (UV) spectrometry is a technique that measures the absorbance of ultraviolet light at 260 nanometres (nm), which correlates with the concentration of nucleotides in a solution. This technique offers a rapid and reliable method for determining the yield of oligonucleotides by evaluating the optical density (OD) of the sample.

PAGE

Polyacrylamide gel electrophoresis (PAGE) is a widely used technique for quality control of oligonucleotide synthesis due to its ability to resolve nucleic acids with single nucleotide precision. At Ella Biotech, we use PAGE for quality control and purification, especially for long oligos where other methods may be limited. While PAGE ensures high purity, it can reduce yield. For dye-labeled oligos, we analyze the results with a fluorescence scanner to detect any byproducts.
